When I try to isolate them, most of them are dead. I don’t really know which requirements are need to keep them alive and/or induce proliferation in those cells.

Are you using any enzymes to digest tissue? You may want to check the enzyme concentration and duration of digestion. You may be dealing with too much enzyme and too long a digestion period. Roche has a selection of enzyme blends that may be helpful.
