One of the main problems about CTCs analysis is their scarcity. A simple method that allows to isolate and possibly to expand these cells by culture would improve our possibility to analyze them specifically.
Hello, how kind of cancer you need to cultivate. We have successful experience with culturing mouse and human patinets CTCs. We use sice-base filtratiom nethod MetaCell.
we want to cultivate colon cancer circulating cells. We plan to isolate them by cytometric cell sorting as CD45 negative cells and then to isolate and to expand tumor cells in specific culture conditions.
Hello, this method will be very complicated. The cells die in sorter but generally the CTCs from gastric or colon cancers growth wery well and quickly - both animal and human.
Hi! Does anyone have experience how to culture CTC after FACS selection of patient derived PBMCs? Is it recommended to use a kind of stem cell medium at the beginning?
Dear Vladimir Bobek- did you culture you CTCs directly from MNCs or following FACS? any idea about the efficiency of the culturing method (% of adherent CTCs)? and did you coat your culture flasks with anything (poly-lysine, fibronectin?) to promote adherence?
