I have the supplement but I need to replace the medium. For what I’ve red, I believe it is DMEM low glucose without phenol red, but does someone know that for sure? Or have use DMEM low as a replacement with success?
The original formulation of DMEM contained 1,000 mg/L of glucose( low glucose). Moreover, DMEM has almost twice the concentration of amino acids and four times the amount of vitamins as EMEM, as well as ferric nitrate, sodium pyruvate, and some supplementary amino acids. DMEM is a basal medium and contains no proteins or growth promoting agents. In general, the red color of the medium due to the phenol red.
My advise will be talk to company technical service person, Since companies do not reveal too much about their kits. But the technical help will certainly resolve your problem since you will be their customer for ever.
I believe the basal medium used is DMEM without phenol red. However, Stempro chondrogenesis/ostheogenesis differentiation basal medium also contains 1 g/L glucose (low glucose), 110 g/L sodium pyruvate and GlutaMAX supplement. See catalog number 10567014 of thermo-fisher scientific containing the requirements similar to the basal medium of the kit.
Thank you very much for your answers. From gibco they told me that it cannot be simply changed for dmem low, I'm going to keep reading, but I think i will have to test it myself in the end. Dmem is very similar and its a common medium for condrogenesis and osteogenesis that require different supplements, so I believe Dmem low it is probably enough.
