We are undergraduate students in biotechnology and we are doing a B-Galactosidase extraction and we wanted to know if ammonium sulfate affects the enzyme in any way at the time of extraction.
Ammonium sulfate affects β-galactosidase depending on its concentration. At Lower concentration (0–20% saturation) it may stabilize the enzyme. Moderate concentration (20–60% saturation) is used to precipitate the enzyme for purification (salting out). But higher concentration (above 60% saturation) could inhibit β-galactosidase activity by altering protein structure or causing aggregation. It could also change the protein's hydration shell and alter its conformation
So, use it carefully, it's helpful in enzyme purification but can affect activity if not removed.
