Most of the interactions between proteins and the surface of the gold is interaction between the cysteine sulphur atoms. If there are important disulfides at the protein surface it is possible that the interaction with the gold will reduce these and disrupt the protein. There is possibly some minor hydrophobic and salt bridges being formed. Overall proteins are considered to remain intact and active when conjugated to gold. Antibodies are often justed mixed with gold nanoparticles for immunogold labelling in TEM.
Hi Sibasish, serum albumin (including BSA) has a single free thiol group and it is important in binding to Au particles. The binding also affects albumin structure.
