It is essential that the solution be completely thawed, and then thoroughly mixed, before removing any of it for use. This is because the solutes separate from the solvent during freezing because they are excluded from the ice crystals. In a partially thawed sample, the solute concentration in the liquid phase is higher than the concentration of the original solution. And, when the solution is thawed, there will be a higher concentration of solute at the bottom than at the top, if the sample is not first mixed. You can see this interesting phenomenon for yourself if you gently freeze-thaw a lightly colored solution several times without mixing it.
Obviously Yes......after thawing on ice....mix them properly before use.....otherwise you will not get the reproducible results...or in most of the cases no results....like in case of BCA...qPCR...
