I have been trying to isolate neurospheres from E13.5-E14 mouse brain (LGE and MGE). Since the are many different protocols in this method, in some protocols, it says that LGE and MGE should be cultured separately. Is it necessary to culture them separately or would it make sense to pool LGE and MGE together to get more neural progenitor and stem cells (or would it harm to culture them together)?
Hi Idil, That really depends on the questions you would like to address. If you are really interested in the differences between MGE and LGE or specific neuronal types, you should do separately. Otherwise, if you just want to get neural stem cell populations, you can pool them up. But in the method parts, you should say the cells were isolated from GE.
