Dear everyone,
I'm culturing THP-1 cells and they seem to be in clusters.
Only one day after changing the medium completely the medium becomes orange and the cells create clusters. I keep them at 2-5X10^5cells/ml, 20ml in T75 flask.
What does it mean? (that they are in clusters) It's a good or a bad sign?

You need to seed cells at a lower density. THP-1 cells are sensitive to cell density. The recommended seeding density is between 3-7 x 10^5 cells/ml. Generally, 5 x10^5 cells /ml is used.
It is not a problem if cells clump as long as they are growing fine and have a normal morphology. You need to remove dead cells if they are present as sometimes these dead cells could be the cause of clumping.
THP-1 cells grow better in conditioned media, therefore when passaging cells, leave 1-2ml of old media and add fresh media to the culture. Do not leave 50% of conditioned media as cells would not have enough nutrients for growth.
