I am planning to work on T2 diabetic animals and check the efficacy of probiotics. My initial plan is to fed B6 animals with HFD for a while. But now, I have the chance to work on ob/ob mice.
My questions are:
1) Are ob/ob mice good models for T2DM?
2) Do I need to fed them with HFD?
3) For a chronic treatment for several mice, are these animals suitable?
4)And for an overall suggestion?

I've worked with both HFD B6 and ob/ob. I also put ob/ob on HFD.
You don't need to feed HFD to ob/ob mice, they are obese and always develop T2DM.
I used to do chronic treatments and it works fine, you might want to adjust your dosages so that injections or gavage volumes are not too high
Just be mindful of other conditions in ob/ob, for example they have a brain of a different size (which needs to be accounted for when doing stereotactic surgery for example).
No need to feed ob/ob mice with HFD. They will develop T2D.
