What do you mean for "equilibrium of the cellular system"?
Ca2+ ions are among the most important second messengers in the intracellular compartment, and a disequilibrium of their physiological concentration can alter many pathways for sure. At the same time, also their extracellular concentration is fundamental for various molecular processes. Anyway, if it may eventually be a problem or not for your experiments depends on what you are looking for.

In general, the excess of Ca2+ ions is toxic for cell and may cause death. In addition, the excess (or depletion) of Ca2+ can cause biochemical imbalance in the cell. You could check if this cell has any type of Ca2+ transporter as a way to avoid this intracellular accumulation.

YES! if the increase of Ca++ concentration inside the cytoplasm is observed/measured during a very long time (significant change in the frequency of the modification of this concentration), because of the mechanisms of regulation of this concentration that exist within all the cells.
