Do C57 mice have to be anesthetized by inhalation before removing the eye to extract blood? Does anyone know the exact anesthesia procedure and is there any relevant literature to recommend? Since the lab is not equipped with an anesthesia machine, is it possible to use the beaker inversion method for anesthesia? Or is there another easy way?
Removing an eye without anesthesia will incite a "stress reaction" in the mouse that might affect your experiment, not to mention the cruelty entailed. The retina and optic nerve are rich in tissue factor, so that removing the eye will likely release tissue factor into systemic circulation. Tissue factor activates factor VII. Removing the eye will also cause a lot of pain, which will exaggerate sympathetic nervous activity that releases von Willebrand Factor into systemic circulation and increase factor VIII activity. The combined activation of factors VII and VIII will sharply exaggerate insoluble fibrin generation in blood and increase blood coagulability.
Absolutly agreed! I'm really surprised that no one at the university I am working at uses anesthesia before removing the eyeball! So we weren't equipped with any anesthesia machines at all! I think it not only affect the results of the experiment definitely, but it's very cruel and inhumane!

Yes, I agree that animal research is riddled with ignorant, inhumane and cruel practices that distort effective research results. You might be interested in the attached article that I published a few years ago. When I was an undergraduate at the Ohio State University I was instructed to use carbon dioxide to "anesthetize" cockroaches for my honors research project, and I was immediately nonplussed by the idea that carbon dioxide has anesthetic properties. It doesn't. It causes a unique form of asphyxiation that mimics general anesthesia, courtesy of its unique chemical property that inhibits the binding of oxygen to the hemoglobin molecule. So, CO2 asphyxiation causes painless brain hypoxia that abolishes consciousness. It's actually quite dangerous. Animal researchers stupidly use CO2 for both anesthesia and euthanasia, which defies logic. You could also use fixed anesthesia with Xylazine 3mg / kg + Ketamine 35mg / kg and Atropine 0.05mg / kg
