Hi everyone, I found some adherent cells in my PBMC culture today.
The PBMC came from a healthy donor, they were cryopreserved before using. These cells are staying in cRPMI (10% HI FBS) with 10 U/mL IL-2 for around 10 days. Today I checked them and I found some adherent and elongated cells present in my culture. I also checked another plate, which I never touch after I thawed them, they show the same thing. Cells in that plate are kept in just cRPMI. All of the cells are in round-bottom 96 well plates.
Can anyone give me some a on it? Thank you!

PBMCs comprise both of lymphocytes and monocytes. Some of these cells (monocytes/macrophages) will adhere to the plate while others (lymphocytes) may be in suspension.
You may want to refer to the article attached below wherein the investigators reported in vitro adhesion and spreading behavior of normal human peripheral blood mononuclear cells (PBMC) isolated by the Ficoll-Hypaque technique.

In fact, the most common procedure for monocyte isolation is by adherence to the substratum after Ficoll-Hypaque isolation of PBMCs. But this method has certain disadvantages such as high lymphocyte contamination and monocyte transient activation.
