I read many papers about exosome isolation from fresh blood or frozen plasma, but not for frozen whole blood. I am wondering whether can I get exosomes from cryopreserved whole blood? One of my workmate told me the blood cells were broken if crypreserved and would affect the isolation of exosomes? What I need is the exosomes and their surface biomarkers. Could anyone give me your prescious advices? thank you so much.
I would think if DMSO is added RBC lysis upon freezing/thawing of sample should be minimal and exosomes can be retrieved specially if storage was at -80 degrees for less than 6 months, at least some exosomes if not more. Try bead pull down methods or immunoprecipitation of target exosome surface protein to concentrate fractions. In my opinion cryopreserved WB should and would work.

You may also try ultra-centrifugation at least on one sample to see if you can retrieve exosomes this way as it is cheaper if you already have an ultracentrifuge. I did that on frozen blood but wasn't looking for exosomes so I can't attest to their quality. Remember to thaw at 37degrees water bath and once liquified move to room temp.
