I am wanting to stain primary mouse cerebellar culutres. I want to distinguish Purkinje neurons from Cerebellar Granule Cells; I was planning to stain PN with calbindin and stain the CGC with Neun. Do you think the Neun will also stain the PN? Is there a more specific stain for CGC? Thanks, in advance, for your help!
NeuN is a Protein which would be expressed in all mature neurons. But if Calbindin is a high specific marker for Purkinje cells the you can do a double labelling with NeuN and Calbindin. Then you can distinguish the calbindin subpupulation from the other neurons. In the attachment you will find an example. In the hippocampal CA2/CA3 region you can see clearly the calbindin positive interneurons and fiber tract which helps to divide CA2 from CA3 region.
