Any suggestions on how to distinguish between activated and exhausted human lymphocytes T CD8 with flow cytometry?
It is known that exhausted lymphocytes T have higher expression of inhibitory receptors, such as PD-1, TIM-3, and CTLA-4; but the problem is activated lymphocytes T can also express those receptors. Is there a cut-off value that can be considered?

In my opinion, you can not determine any cut-off for distinguishing these cells. because they are not distinct cells population, they are the same cells in different phases. Change in the expression of markers is gradual. however some publications of introduced CD127 and CD62l to distinguish activated and exhausted cells or others pointed out CD69 and HLADR as markers for early and late activation phases, It better to distinguish them through their function such as cytokine production or lysis capacity. Functional tests are more valid.
