Dear Madam/Sir? Can you clarify my doubt? When I raised the polyclonal antibody in Rabbit against a cucurbitaceous host virus and was evaluating it against the infected leaf samples of N.benthamiana , there was difference in the O.D values between healthy and Infected N.benthamiana, but it is not 2 fold or more than that to consider it as a positive. Generally we take a 2 fold difference between a healthy and infected samples to consider as positive. At that point of time, I was not having infected cucurbitaceous hosts to validate the antibody. Later, when i had the infected samples of cucurbitaceous hosts such as muskmelon, bottlegourd, cucumber and validated against those there was even upto five fold difference between the infected and healthy samples. Is it because the virus multiplies luxuriantly in a cucurbitaceous hosts compared to model plant N.benthamiana which belongs to solanaceae . Or is it because of anyother factors that is responsible for lower reading in N.benthamiana. Can you furnish your ideas or opinion?
