Saurabh, most animal mtDNAs have no introns.

The primers should work, just be carefull with the location, because polycistronic transcripts are cleaved.

I wanted to update you that primers designed for mitochondrial genes (DNA) do work for cDNA from mitochondrial transcripts. As suggested, I checked that the sequence should not be in a region between two exon coding sequences (since mitochondrial transcripts are polycistronic).
