I tried to measure IL-10 and IL-1b secreted by HT-29 cells but I couldn't detect by ELISA. Literatures say that it can produce them. Has anybody ever measured IL-10 from HT-29 carcinoma intestinal epithelial cells?

Did your standard curve work? Another option is to "spike" your samples with a known amount of each cytokine you are testing so that you know how sensitive the Elisa is.

I didnt dilute my samples. Is it possible that something in the media interfere with the connection of IL-10 to anti-IL-10?
One plate is for IL10 and the other with darker standard is for IL1beta.
None of them were positive (HT-29 stimulated by LPS supernatant)
You have some signal in rows 1 and 2, thought it is very week. Do you have the culture conditions, including how supernatants were harvested.
Yes. Imagine two first rows
Each row has 12 wells. Four first rows were control four second were lps four third were my component that i expect to increase il10
But elisa reader detected the four third wells as negative
