We use isofluorane anesthesia when we do intranasal administration in mice. Volume has a strong effect on where the inoculum ends up. We use two inoculation volumes, 2ul for delivery to the upper airway, and 20ul for delivery to the lower airway. Anesthesia is absolutely necessary for the higher volume. However, we use it for all i.n. procedures to minimize stress.
Further, it helps to watch the breathing of the mouse for a few breaths to time your administration during inhalation.

I have done also in the past intranasal admnistration in mice. You can anesthesize mice by intramuscular injection of 150µl of a solution containing 10 µg/ml Ketamine and 10 µg/ml Xylazine. I have the same doubt, because many papers describe infections or one shot treatment by intranasal route using anaesthesia, but in case of a daily treatment, I'm not sure if it's allow to apply anaesthesia everyday. Is somebody facing the same problem?
