I am struggling using cyclic nucleotides from InVivogen (c-di-AMP, and 2'3'-cGAMP) on my THP1 cells.
I use the nucleotides directly on my cells and I have really weird results (high EC 50, cells respond better if they are just thawed = cell permability can be different). In some publications, I read that the authors used permeabilised cells or even transfected cells (Lipofectamine...) (not mentioned on InVivogene website of course).
I am not sure if the nucleotides can go through the cell membrane by their owns? And of course the company developed cyclic nucleotide with the incorporation of fluorine which, apparently, increases the lipophilic properties/efficiency of the compound (so can across the membrane).
Do you have any idea?
