Dear All
I want to label in-vitro transcribed RNA with Cy5 dye on one end, what could be the best possible way to do that. For DNA, I am simply labelling one of my Primer with Cy5. If I label Cy5 on the 5'-end upstream of T7 promoter on a primer, would that work, if not, what could be the best possible way?

Thank you for this reference. I also came to the same approach, the click chemistry based approach. I found out that this can be done in vitro as well. Jena Biosciences provide the kit. It is very interesting approach where you incorporate Azide-labelled UTPs during your in vitro transcription and then treat the labelled RNA with DBCO which contains Cy5. The Cy5 molecules bind to the Azide and thereby giving you the labelled RNA.
