Hi! I'm performing some native page but I'm having problem. During the pre-runnin the gel is ok, but when I load my samples and start the second running, at the bottom of the gel appears a strange line, like if there are cristals. Does it happen to someone else? Thanks!
Hello Valeria. What buffering system are you using? And, are all of your buffers made up in Grade 1 (MilliQ-grade, or equivalent) water?
Running buffer 1x made with Tris base and glycine for the anode side and running buffer with 1% DOC for the cathode side. Usually we left the buffer and the gel at RT O.N. before running. All the buffer are made up with MilliQ water
It sounds weird something would happen at the bottom of the gel, if you loaded your samples at the top of the gel.
Is the same happening, if you load only loading buffer or nothing at all?
Yes, it happens also in lane without sample. It seems something indipendent from the samples since this line is horizontal compared to the lane of the sample
