I'm wondering if anybody here has used CRISPR in bacteria (Vibrio/Pseudomonas) to delete large (>10 kb) chunks of DNA. I appreciate any pointers/resources/commercial kits.
We did not do it yet, but for sure we are going to try it next year. I have a project about P. aeruginosa and it's resistance and we are going to try delate some parts of it's genome. So, please be in touch if your are interested, and we will see how it is developping.

I found an article where they've done it using nickases instead of the typical dsDNA breakage (DOI: 10.1021/acssynbio.5b00132). Is your planned approach similar?
