Has anyone been successful in amplifying the sodium-potassium-two chloride transporter in caridean shrimp gills? Could you provide the primer sequences please.
Hi John, as I can't provide you the primer sequences directly but i can give the ways to design a primer sequences...
first, go to "PubMed" homepage, you can see the search bar and then in the lefe drop-down list choose " Nucleotide" and then type the primer name like this " mouse or human , primer name"in search bar , you will find so many results, click one you think may possible , in the " ORIGIN" section ou can see many sequences, copy one of them then paste to "http://primer3.ut.ee/" (a primer design website) ,
Select the Task for primer selection: pick_primer_list
go to "General Primer Picking Conditions" section,
Primer size min: 18 max: 25
Primer Tm: Min : 58 Max : 62
Primer GC%: Min : 40 Max: 60
at the bottom of this section, click the button of "pick primers"
the result of left_ primer and right_primer sequence is what you find...
And another way to get a primer sequence is referring to previous studies, copy it directly...
Thanks Huan. We have developed successful primers for this transporter in several crab species, but we are not having much success with caridean shrimps. I'll see if anything comes up in PubMed then. J.
