We are using a Klenow fragment like enzyme in qPCR assays with Taqman probes. Contrary to expectations, we obtain good amplification curves, despite the enzyme lacks 5'-3' exonuclease activity. So, the probes are actually being hydrolized. How can this be possible? Residual activity? Have you ever experienced that?
