I'm looking for apply GreenTaq Master Mix in order to detect bacterial strain fingerprintings using (GTG)5-PCR and BOX-PCR. It is possible to apply it?
I've worked with BOX-PCR before, and it doesn't matter abt which Taq you are using. It's different from a conventional PCR just in their theory, primers and the thermocycling parameters.
I think the matter is different here. In conventional PCR Master mix can be used freely for amplification of bacterial DNA but, in Box-PCR , I think no.
In human for example, in fingerprinting we used Real Time PCR for amplification of STR using Promega kit.
This, I advise you to follow Promega company products. Kind regard
You can use it, although result can be not the best. rep-PCR usually need optimization of all components of PCR mix (dNTPs, Mg concentration, etc.) to get repeatable profiles.
