Measure the haematocrit or the RBC concentration of your blood sample, so that you can express your result based on Mio erythrocytes or something like that.
Then, I'd prepare red cell ghosts by hypotonic lysis of RBCs followed by centrifugation. You can now measure protein concentration by, say, BCA or Lowry assay.
Phospholipids can be determined by phosphate measurement after ashing with nitric acid:

To measure any particular protein like actin or band 3 will probably require immunoassays like ELISA. If quick-and-dirty results suffice, SDS-PAGE followed by scanning of the stained gels may also work (that depends on your question).
