Sample preparation will depend on the type of drug and cell matrix. Organic solvents, acid treatment, sonication etc can lyse the cells and get intracellular content for quantifying the drugs being taken inside the cell. Before you do that you have to check the matrix effect. You would require to remove proteins and other unwanted contents to reduce matrix effect for which you would require wither solid phase extraction, liquid-liquid extraction, syringe filtration etc. For this type of study an internal standard greatly help to calculate the recovery from matrix and also in data normalization. But you have to be more specific to get a better answer. In short, there is not a universal protocol for this which will work for all analytes in all cell lines/biological samples.
