Hi, I wanted to make a colon cancer in mice to see the effect of 2nd generation mTOR inhibitor. As, I have no experience on this, which would be the best cell line and mice model to induce. Need your valuable suggestions.
If you are interested in using a human cancer cell line here (and not a syngeneic model) you will need to use an immunocompromised mouse model - NOD/SCID or nude mice are commonly used strains for colon cancer in vivo studies.
SW480 cells are tumorigenic in mice and could be useful, however there is lots of literature out there which may be useful in designing your experiment.
I guess HCT-116 cell line would a good choice to study colon cancer. Moreover, if you are interested in the In vivo xenograft studies, I think BALB/c nude mice would be the best model. Perhaps, the below-mentioned link can help you in a better way.

Actually, I'm planned to select LS174T cell line (as it is also pretty well for developing tumor xenograft model) and athymic nude mice as a cancer model. As I'm beginner in the field of cell culture, one of my senior lab mate said it is easier to handle LS174T cell line than the SW480. What you guys suggest about the LS174T cell lines and Athymic nude mice? TIA.
Hello, for my experience the best cell lines I used were HCT-15, Colo320, and HT-29 all of them worked fine in xenograft studies. They produce tumor without any problem. You can use Nude or Scid immunocompromised mouse model for this kind of xenograft.
Dear all, I am planning to inject SW480 in nude mice for in vivo studies. please recommend the injection cell numbers (exact number per mouse) and how long the tumours became palpable and when do the tumour reach the end point of 1500 mm3. thanks heaps!
