I want to determine the lipid profile of fish liver and muscle but I am not getting any suitable and simple protocol. Could anybody suggest some simple methodology?
sir, it is for only total lipids or i can do complete lipid profile by this technique. What about enzymatic methods (commercial kits available in market).
Folch method is for lipid extraction, then use gas chromatography for fatty acid profile, reverse phase HPLC for TAG and DSC for thermal profile.........
I have no experience working with the commercial kits........
Hi, unfortunately the full Lipidome of liver includes a multiplicity of lipid species and won´t be simple. All in all you can try either acidic Folch (low pH for quantitative extraction of free fatty acids) or Bligh and Dyer which should extract you most of the lipid species. If you just want the FA composition of the total lipid fraction you can to the transmethylation coupled to GC/MS (or GC/FID). If you want to have a differential pattern of liver and muscle lipids you can either perform qualitative thin layer chromatography which gives you an idea about the differences or you have to perform LC/MS. Anyhow LC/MS will show you that there are plenty of different lipid classes and subspecies out there so it is very important to know which lipids are of interest.
