I am working with primary rat neuronal cultures and am looking to inhibit caspase-3. Many of the inhibitors available also inhibit a number of other caspases but I am looking for specificity.
Years ago I worked with DEVD-CHO in primary hippocampal cells. Retrospectively, I am aware that this is a "dirty drug" regarding its specificity for caspase-3. An alternative compound from AstraZeneca seems to have remarkable specificity for caspase-3 over other caspases along with a reasonably low toxicity. It is called AZ10417808 and distributed by Tocris Biochemicals. I personally have never worked with AZ10417808, but it may be a good solution for your in vitro experiments?
