I am currently extracting DNA from plasma samples of HBV-positive clinical cases using the QIAGEN Mini Kit (250), following the manufacturer’s protocol step by step. Despite adhering strictly to the procedure from lysis to elution I consistently obtain very low DNA concentrations, as verified by both Nanodrop and fluorescence-based assays.

I have experimented with elution volumes of 200 µL and 100 µL, but the yield remains low. Could this issue be related to the performance or compatibility of the kit with my sample type? Alternatively, should I consider switching to a different kit or extraction method?

Any suggestions on optimizing the protocol or recommendations for alternative kits better suited for HBV DNA extraction from plasma would be greatly appreciated.
