Hi folks,

I have been trying to co-transfect plasmid and siRNA in HeLa by using reverse transfection with Lipofectamine 2000 (L2K) in 96 well plate for a reporter (psiCHECK-2) based screen. I have done some initial optimisations. I have realised that I need to use less volume (0.3uL/well) of L2K in reverse transfection than forward. After I optimised cell number (25,000) and L2K volume (0.3uL/well) using the reporter plasmid, I decided to add siRNA targeting renilla. Whenever I combine, the assay (luciferase assay) doesn't work. I do not get firefly signal. What could be the reason? Have you ever done any similar experiment before? Could you share your experience? Thanks
