Hi,
I am working on stable cell lines of MCF-7 cells overexpressing different truncations of my gene of interest. I Found difference in rate of proliferation between these cell lines . However, when I checked the cell cycle kinetics by PI staining, I did not find any change in cell cycle profile between these stable cell lines. I was expecting that the cell cycle progression also corelates with my proliferation data. Is it possible not to see any difference in cell cycle progression although there is difference in proliferation rate . How to explain this data ?

This difference is possible if the cells are in different stages of the cell cycle, or some cells have been stopped at a certain stage of the cell cycle, for example, as a result of exposure to growth factors or cytokines.

Maybe the changes that you are noticing are related to cells detaching/dying rather than changing its proliferation rate.
Maybe the truncated versions of your gene of interest make your cells more or less susceptible to certain types of stress.
How are you measuring proliferation? Which are the corresponding proliferation rates for each of your clones?
