It is according to which cell you will use. For 293T ,CMV vector is ok to express your mammalian antibody

Thank you for your advice. Yes, you do have a point about the CMV vector since it has a strong promoter for high level expression. However, I was thinking about a vector that can express the 2 chains. When using the CMV transient vectors, I will have to do a 2 plasmid transfection for the light and heavy chains respectively.
I would be happy to listen to your advice or correction.

I used the expression vectors from InVivogen (pFUSE...) for recombinant mouse IgG. I also subcloned the antibody genes into vectors for lentiviral transduction to achieve stable expression. It worked well but the IgG amounts, which were produced, were not so high. But the reason was probably my target cell line, which I used for the lentiviral transduction.

I have used pFUSE for cloning an Fc-fusion protein and it has worked well.In fact, this was the best vector that worked with these types of constructs in terms of expression. I have never tried it with a VL-VH sequence but I guess it should work the same way. A lentiviral transduction system should be useful for stable expression.
