Dear Colleagues,
I need specific anti-Rac1 antibody for immunofluorescence staining. I've tried two up to know (Cytoskeleton and Thermo), but the staining is predominantly nuclear or some unspecific dots. I can't see any accumulation of the signal in the lamellipodium nor after FBS deprivation and EGF stimulation. Also I tried to validate the procedure by transfecting the cells with GFP-tagged Rac1 and doing the staining afterwards, but the signals were completely different.
We routinely perform immunofluorescence in the lab and I am pretty sure that the protocols and secondary antibodies are working ok. I also performed some optimization of fixation, blocking and dilutions of the tested antibodies, and even contacted the support, but with no results.
I would be grateful for any suggestions.

I'd recommend doing a search on BenchSci (https://www.benchsci.com). There are a couple options from BD and EMD that have been published. Please see screenshot below.
It might be best for you to review the published data before deciding which one to purchase.

You can have a look at https://www.labome.com/review/gene/human/Rac1-antibody.html . A couple of monoclonals have been validated in knockout samples. At the webpage, search for 'IC' will lead you to the antibodies that have been used for immunocytochemistry. It looks like BD 102/Rac1 clone https://www.labome.com/product/BD-Biosciences/610651.html might be a good fit for you. You can also follow the links to view the actual articles.
