I would like to fragment genomic DNA and cut out a gene of interest with restriction enzymes. To do so, I need a set of enzymes that cuts up- and downstream of my gene, but not within it. For further fragmentation of the gDNA, I will need some more enzymes that don't cut within my gene. Ideally I would like to use high fidelity enzymes.

Can you recommend a tool that allows you to enter the sequence you want to cut (in my case, sequences up- and downstream of my gene), but also exclude enzymes that cut within a certain sequence (my gene of interest)?
