I have a question regarding the detection of lipolytic activity. If you mixed olive oil in an agar culture medium, would you be able to observe halos around colonies that have a lipolytic activity, or would you need another method?
Of course you will observe the halos on olive oil-minimal agar medium. I call it precipitation zone because that is what the halo looks like around positive lipolytic bacterial colonies. The halos are better with Tween 20 or 80 but nest with tributyrin agar and spirit blue agar. Addition of rhodamine B accentuates the halos and make for more ready observation and measurement.
