I'm isolating genomic DNA from human saliva that was frozen but not stored in Oragene solution (it was previously used for other analyses). Our yields from ~0.5 - 1ml of saliva are extremely low and of poor quality, due (I think) to enzyme degradation. Any advice for increasing yields/quality? We are currently using Qiagen DNA micro kits for column-based isolation.

Zymo Research consider good kits bodily fluids.and you can extract using the Wizard Genomic DNA Purification Kit (Promega), getting a 40 microliters of a DNA with a concentration of 100 micrograms/microliters.

You now the Yield depend on the quantity of whole saliva samples , and if we use 300microliter of blood the yield be 5-15 microgram of DNA , in saliva the best procedure in link studies.
