I am using chitosan to modify the surface of biochar. 3g biochar is aaded into 200ml gel of chitosan (2% w/v). Then the mixture is soaked in NaOH and and then washed by DI water. But I do not know how many chitosan is loaded in surface of biochar. Please help me for that.

Do you have a spectroscopic method like UV or such to evaluate concentration of chitosan in solution? From known qty that you start with, all that remains in the filtrate. NaOH washings, and DI washings get complexed qty by difference.
However if desired effect is to have chitosan active sites for further ion complexation, then use a known concentration of that ion ( say Fe +3) and see how much is left in the solution after immobilization to the chtosan biochar complex.Untreated Biochar would have to be run under same conditions as control and determine difference. A chitosan ion complex to determine maximum loading per gram of soluble chitosan could be used to estimate the chitosan content in the biochar complex.
Ion microprobe of the biochar chitosan complex scanning for the metal ion of Fe could be used to determine the coverage also directly.
