I'm working on a cross streak assay with P. chrysogenum and S. griseus, however culturing S. griseus has been difficult even when I have a pure stock culture. The stock tube has descriptions for proliferating the bacteria, but I wouldn't mind additional direction.

in general S. griseus should'nt be hard to culture. Is your strain a type culture strain or wild one?
Usually, streptomycetes grow well on Starch Casein Agar (also works fine with Funghi), Carbon Utilisation Agar, Actinomycete Isolation Agar (with some glycerol), Tryptone Soy Agar and many other media for general growth. Medium does'nt need to be rich, especially if you would like to induce some secondary metabolism reaction (they often need to be stressed to fight).

I've been culturing S. griseus from a type culture from Ward's Science. I've tried several agar varieties, but I may not be giving Streptomyces enough incubation time. Can I get your opinion on how many days, I've tried (3-7 days), a Quad Strk. of S. griseus would take at room temperature? So, your suggestion for inducing secondary metabolite production is conditioning them to a minimal media? I intended to perform the cross streaks on ISP2 (YEME), but you advise against such if I'm looking for antibiotic production? Can you refer others ways of inducing antibiosis?

For my S. griseus (ATCC 10137) 5 days at room temperature is enough for the strain to produce spores. Production of secondary metabolites in streptomycetes is the most active while the bacterium switches from substrate pseudomycelium to aerial one (which ends up in producing spores). In order to produce aerial pseudomycelium they need to be stressed, where nutrient stress is quite effective and has been linked to production of antibiotics. I think that YEME is better for biomass production but not necessarily for antibiotics secretion. Among other methods, one the most known is competition mediated production (when you culture streptomycete e.g. with mould).Cheers.
