I notice that most people use 4-OH tamoxifen for IP administration and tamoxifen citrate for oral administration. Tamoxifen citrate needs to be metabolised by the liver to be effective so I assume that it should work via IP. However, I cant see that anyone has used it.

I have not used tamoxifen in vivo, but I guess it should work when administered IP. However, as you say, 4-OH tamoxifen will not need to be metabolized by the liver, so you may reduce the variability between individuals by using 4-OH tamoxifen. In humans, genetic variants in CYP2D6 lead to variable metabolism of tamoxifen in 4-OH tamoxifen, hence in treatment efficacy.
I have a relevant question. I want to induce conditional knockout in pancreatic beta cells. I fed my mice on Tamoxifen containing diet starting from 4 weeks of age for 1 month as this tamoxifen administration protocol is established in my lab and works with other cre in other tissues but it didn't work for my system.I am thinking of giving tamoxifen as intraperitoneal injection. Do you think it might work better?
I just had the same question why most references are using tamoxifen in oil IP injection rather than Tamoxifen citrate IP injection.
even oral garage can be with tamoxifen citrate too?!
