Is there instructions for VTM centrifugation before proceeding RNA extraction?
Since the VTM swabs contains 2-3ml and commercial Coronavirus PCR kits LOD about 100 copy/ml What is the best preparation for VTM before extraction?

I think the best way to address this question is for you to directly try it in some of your samples, especially the confirmed RT-qPCR positive ones by whatever product you are using. You can do head-on comparisons wherein a sample will be divided to RPMI, VTM, UTM and the transport reagent that you are using to check.
SARS-CoV-2 is more resilient in samples than most people give them credit for, especially if the time period from sample collection to RT-qPCR testing within 24 hours. Dear Hazem Almasri There are many ways to prepare in-house VTM for sample collection. The Centers for Disease Control and Prevention (CDC) last year put up a document for the preparation of Viral Transport Medium which majorly requires:
- Hanks Balanced Salt Solution (HBSS) 1X with calcium and magnesium ions
- fetal bovine serum (FBS)
- Gentamicin sulfate (50mg/mL)
- Amphotericin B (250ug/mL) and few more things.
So yes, it seems logical that you can use RPMI/ EMEM supplemented with FBS and an appropriate amount of antibiotics for the same purpose.
PBS also you can use for nasopharyngeal swabs storage media instant of VTM.
