Dear All, mirna primer showing some problem in the melting curve? any idea why? As attached is the melting curve. The forward sequence is obtained from miRBase and reverse primer is universal.

Dear Saliha,

Circulating miRNA is presumably not naked miRNA, which would be degraded within seconds due to the high levels of nucleases in blood. Several reports have demonstrated that stability results from the formation of complexes between circulating miRNA and specific proteins or been protected in the exosomes or vesicles.

With this I think that keeping the blood for one day before preparing mirRNA will be ok.
