I have been noticing increased differentiation in my mouse embryonic stem cell culture. We culture cells in both 15%FBS.KODMEM media as well as the the 2i /N2B27 media. For the serum containing cultures, we use 1000U/ml of recombinant LIF. For contest we are comparing phenotypes between the two media conditions that we have observed, only there is greater differentiation in serum cultures (as expected) and increasing LIF to about 2000U/ml seems to do the trick! But I'm concerned if this could be affecting the cell dynamics in some unknown manner. Does anyone have opinions about this?
