Dear all,
I would like to cultivate Lactobacillus acidophilus from the L-dried sample. I do not have any experiences in culturing bacteria other than E. coli and my lab has no MRS media. Could somebody suggest if Lactobacillus acidophilus can be grown in LB media just like E. coli. The organization mentioned MRS media for revival and cultivation in the invoice.

While I can't speak for your particular strain, there is a principle: Usually, the media don't create perfect stringency, but rather they bias the growth of one organism over others. On MRS, Lactobacillus outgrows all the others. So even if other bugs fall into your dish -- which they will, unless you work in a GMP grade clean room -- they will be outcompeted by lacto.
When you use suboptimal media, that's no longer the case. It creates a huge risk that your lacto creature will be overgrown by contaminants and lost. So it's usually fine in the short run, but if you care for the long term health and purity of the culture, I'd go with the recommended medium.
I recommend you MRS medium. Some strains of L. acidophilus are a little hard to cultivate
You can use MRS for the growth of your Lactobacillus and incubate it aerobically, LB is not appropriate for LAB because it lacks some essential nutrients, the pH influences too.

Thank you very much for your suggestions and comments. As suggested, I will strictly use MRS media for the growth culture. However, as I do not have facility for aerobic growth, I will try to use para-film wrapped plates to attain partial anaerobic condition.

Hi Bikash,
From what I understand, this bacteria is microaerophilic. Usually 5% carbon dioxide is required for growth. I am not too sure if parafilm would help to create this environment.
