Hi everyone.. is it possible to analyse our data saved as JPEG in ImageJ? I did tumourspheres culture under hypoxic condition and have to take the pictures of my spheres in multiple 6 well plates, which usually took quite a lot of time and I try as much as possible to minimise the time the plates are outside due to the hypoxic condition. So usually I saved my picture in JPEG without the TIF format. I knew that usually analyses are usually done on TIF-formated picture but just wondering if anyone have the experience to analyse ImageJ data using JPEG image? And how do we measure, say the diameter or area of our cells using ImageJ?
ImageJ can read many file formats. Analysis is done after file reading, and does not depend from format.
For your task you can use commands:
- Image - Adjust - Threshold,
- Analyze - Analyze Particles.

I agree with Alexander, ImageJ can read many file formats including JPEG.
initially you will get the unit of picture in pixel so you have to set scale.
Calibrate slide (micrometer measurement slide) required to set scale and you have take the picture in same magnification, same microscope and same camera. after you set the scale you can measure anything in your new unit measurement

The tiff is used mostly for multispectral images, other than that it has nothing so special.
If your files do not have any multispectral aspects, you can perform everything with .jpeg format as well. Nevertheless, it would be better for you to record the files in .png or similar formats which support lossless compression and containment.
