Our embedder is consistently overheating the wax to around 70C and we are unsure if this will damage the tissue for later IHC or make the block unsectionable.
If you could provide any papers that investigate this that would be great!
It depends on the antibodies. For some of them high temperature is critical but on the other hand alot of comercialy available antibodies need antigene retrievel. That meens water bath or microvave treatment in citrate buffer ph 6 around 100°C. May be you will need such pretreatment as well. The blocks are sectionable when they are properly dehydrated. So try it!
The problem could be with the paraffin itself. 70 degrees is borderline. Overheating the paraffin can cause it to break down and make slides difficult to obtain. There should be a melting point temperature on your paraffin and the embedding temperature should not be more than 10 degrees more.
