In my group we are planning to use an ELISA kit in order to measure VEGF in serum from relapse multiple myeloma patients. We wonder whether we could have problems with the peak in immunoglobulin we expect at that point in our patients, in term of interference with the measurement of VEGF. Does anyone use this kind of kit in this sort of patient?

My experience is that levels of Immunoglobulins (Igs) are only a problem in case of measuring Igs with anti-human Ig antibodies for detection (e.g. indirect ELISAs) - thus unspecific binding of Igs to ELISA plates (e.g. maxisorp) will cause a false signal. Your detecting antibody should not react with Igs I suppose.
To be absolutely certain (in case you have suspicion of false positive signals due to unspecific binding of Igs to the plate) you could spike additional Igs into a test sample or precipitate Igs – in order to see if this affects measurements.
Thank you Dres, that is what I had in mind, I expect the antibody in my ELISA kit will be specific enough to avoid the binding with Igs.
