FBS contains a lot of factors and proteins that will behave as foreign molecule in the mice so i think it should not be used. u can use PBS or Medium without FBS.

After tumor formation, going to any experimental you should use PBS or Medium without FBS. So, Yokesh said answer is correct. If you are working BALB/C nude mice u should follow accordingly our statement.
Formally speaking you would have to compare FBS alone to your Conditioned Media. Still, I would try to avoid that because FBS itself will have difficult to control and interpret effects due to the multitude of factors present as well as batch-to-batch variations. It could be very difficult to repeat an experimental finding because of that. There is a nice paper by Bob Dickson, Marc Lippman et al 1986 in Science Vol 232:1450-1543 that describes an experiment with conditioned media in nude mice with tumors. Would follow the approach described there and attach a pdf of the paper.
Thank you all for the reply. Actually I have the conditioned media that is prepared by growing some specific cell type in presence of FBS without which cells may not secrete essential factors or factors may be unstable.
Thus if I have to see the effect of the condition media then what is the alternative way out?
Aditya, what is the purpose of using conditioned/complete medium in vitro? Because we need to have proper atmosphere that is pretty much similar (or at least closer) to the real environment in the human body. That is, if you inject nude mice s.c. using matrigel + your cells that is suspended with the matrigel in normal saline then all your cells will stick together and not diffuse, that's at first instant. Second, they should be growing fine if these cells are cancerous cells (for example).
You have not mentioned the type of cells that you're trying to use and what is the origin of these cells. Not every element is suitable to be injected into mice. Always remember, mice are living animals and their body will respond to changes accordingly. In addition, adhere to the welfare of the animal, as you don't want to put anything that you think it may or may not work in animals. Preparing and designing proper model is very essential for experimental outcomes.
